Extra-virgin olive oil sets itself apart from other plant-based oils in the same price range largely because of its polyphenols. Among those compounds, the two most frequently cited in the scientific literature are oleocanthal and hydroxytyrosol. Understanding what each one actually is—and where the evidence begins and ends—gives the word polyphenol the weight it deserves.

Hydroxytyrosol and the Narrow Recognition from EFSA

Hydroxytyrosol is one of the defining phenolic compounds in EVOO. Following an assessment by the European Food Safety Authority (EFSA), EU Regulation EC 432/2012 permits producers to carry the claim that "olive oil polyphenols contribute to the protection of blood lipids from oxidative stress"—but only when the oil provides at least 5 mg of hydroxytyrosol and its derivatives per 20 g of oil. That threshold translates to a polyphenol content of 250 mg/kg. The figure has since become the de facto benchmark against which EVOO polyphenol levels are measured across the industry.

The Signal from Oleocanthal

Oleocanthal is considered one of the primary compounds behind the distinctive peppery catch at the back of the throat that characterises a fresh, well-made EVOO. A study published in Nature in 2005, conducted by researchers at the Monell Chemical Senses Center, reported that oleocanthal inhibited COX enzymes in vitro in a manner similar to the non-steroidal anti-inflammatory drug ibuprofen. That said, the finding was a pharmacological observation at the test-tube level. It is too early to conclude that the same effect occurs at clinically meaningful levels from the amounts of EVOO a person typically consumes in daily life.

Where the Claims Outrun the Evidence

Phrases such as "oleocanthal is a natural painkiller" or "hydroxytyrosol stops ageing" are not conclusions that current clinical research supports. Regulatory guidance in multiple jurisdictions restricts definitive efficacy language on food labels for precisely this reason. There is also a practical dimension: polyphenol concentrations fall quickly after pressing, accelerated by light, heat, and the passage of time. A number printed on a label is a snapshot, not a permanent truth about every pour.

The Place of a Single Bottle

Polyphenol figures only carry meaning when freshness is still intact. The recommended storage conditions are a dark location at 14–18 °C; once opened, finishing the bottle within 2–3 months is the most sensible approach.
